Sleeper Floor or alternatives

I'm having a builder add a kitchen addition. It will be slab on grade and is located in the mid-Atlantic area.
What is the best way to control moisture and avoid issues with mold and rot. my builder proposed the standard approach of 6mil vapor barrier under the concrete and then 2x4's and then plywood subfloor. While this has been the standard way, I'm wondering if there is a better approach or what materials I should look out for. I've already asked him to upgrade the subfloor to 3/4 Advantech T&G. The final floor will be solid oak hardwood.
